Gram differentiation was obtained with 95% (v/v) ethanol in water, this was found to extract much less of the dye iodine complex from intact bacteria than from the lysozyme-depolymerized forms. The latter lost about 80–90% of their net dye and iodine content, whereas intact bacilli lost only 20–45%.

Bacillus megaterium is a rod-like, Gram-positive, mainly aerobic, spore forming bacterium found in widely diverse habitats. [ 1 ] [ 2 ] It has a cell length up to 100 μm and a diameter of 0.1 μm, which is quite large for bacteria. [ 3 ]
7 Οκτ 2024 · Exploring the staining techniques for Bacillus megaterium provides valuable insights into identifying and characterizing Gram-positive bacteria. One of the most commonly used methods is the Gram staining procedure, which differentiates bacteria based on the composition of their cell walls.
26 Ιουλ 2007 · B. megaterium is a Gram-positive, mainly aerobic spore-forming bacterium found in widely diverse habitats from soil to seawater, sediment, rice paddies, honey, fish, and dried food. It can grow in simple media on more than 62 carbon sources out of 95 tested, including all tricarboxylic acid cycle intermediates, formate, and acetate.

A comparison of the roles of the cell wall, cytoplasmic membrane, and cytop lasmic components revealed that the intact cell wall was the dominant contributor to the gram-positive state of Bacillus megaterium.
