IL11RA requires the coexpression of gp130 receptor common subunit for signal trans-duction (Nandurkar et al., 1996). The major pathways that are activated upon IL-11 stimulation are JAK-STAT, Ras-MAPK, PI3K-AKT, and NF-kappa B (Du & Williams, 1997; Li et al., 2012).

Structural shifts in IL-11 Mutein underlie inhibitory activity by altering cytokine binding interactions at all three receptor-engaging sites and abrogating the final gp130 binding step.
