Runite ore can be somewhat profitable to mine since it is used to make rune equipment and elder rune bars. There's a few places you can mine runite ore but one of the best is in the Mining Guild beneath Falador.
If you hover over ore types in mining menu, it gives you a list of locations where you can find each ore. There are some at Crandor or the Karamja volcano mine. If you have 60 Mining, Mining Guild has runite rocks, amongst many others. Tip: if you go to the in-game mining skillguide, click on the exclamation mark behind any ore (e.g. runite).
At level 50, the only locations available are inside Karamja's volcano, Crandor Island and just north of Edgeville in the wilderness. Not entirely close / safe to pick. The mining guild has the best access to runite rocks, but requires 60 mining.
By doing quests and unlocks, more locations to mine runite will become available to you. Non-members can only mine runite at one location - the Wilderness Rune Mine, which contains two rocks. It is located north-west of the Lava Maze in level 46 Wilderness.
Mines are places and areas where various minable resources (e.g. ores, essences, stones, gems, etc.) can be mined. These may be above ground (quarries or outcrops) or underground (mines) and are scattered all over Gielinor.
Runite ore can be mined at level 85 Mining providing 125 Mining experience. After runite rocks are mined, it will take twelve minutes until it reappears. Runite ore is used with eight coal to smelt runite bars. Smelting a runite bar gives 50 Smithing experience.
