DLL files might require other DLL files. In the same way that an application requires a DLL file, a DLL file might be dependent on other DLL files itself. If one of these DLL files in the chain of dependency is not found, the application will not load. This is debugged easily using any dependency walker tools, like Dependency Walker.

Although .EXE files and .DLL files are essentially identical in terms of format, the distinguishing feature of an .EXE is that it contains a designated "entry point" to go and do the thing the EXE was created to do.

Often times a developer on my team will create a new Visual Studio project and reference a DLL somewhere on their local machine (e.g., C:\mydlls\homersimpson\test.dll). Then, when I get the project from the source control repository, I cannot build the project because I do not have the referenced dll in the exact same location on my machine.
The only DLL files that I know of that work natively on Linux are compiled with Mono. If someone gave you a proprietary binary library to code against, you should verify it's compiled for the target architecture (nothing like trying to use am ARM binary on an x86 system) and that it's compiled for Linux.
