Populations of cells can be analyzed biochemically by disrupting them and fractionating their contents by ultracentrifugation. Further fractionations allow functional cell-free systems to be developed; such systems are required to determine the molecular details of complex cellular processes.
In this chapter, we describe a simple and efficient method to isolate the different compartments of Escherichia coli by a fractionation method and to determine the presence of the protein of interest. For inner membrane proteins, we propose a method to discriminate between integral and peripheral membrane proteins.
METHODS TO DISRUPT CELLS. After the material to fractionate has been chosen, the cells need to be disrupted. The disruption method needs to be suf ficient to break open the cells but not so disruptive that the organelle or fraction to be isolated becomes overly damaged and/or nonrecoverable.
