INTRODUCTION. Most environments carry a mixed microbial population. To fully appreciate the contribution of each group of organisms to the ecology of the mass, one must first dissect this mixed culture to obtain single colonies.
This guide contains general technical information for bacterial growth, propagation, preservation, and application. Additional informa-tion on bacterial culturing can be found in Bergey’s Manual of Systematic Bacteriology, 2nd Edition, published by Springer, New York.1.
This chapter talks about the main culture techniques for microbial growth. It first describes the solid, semisolid, biphasic, membrane surface, and immobilized culture techniques. This is followed by laboratory scale liquid culture techniques including specialized liquid cultures such as synchronous and dialysis cultures.
