In most protocols, sections are stained with a standard alcian blue (pH 2.5) method followed by PAS. The alcian blue stains sialomucins, sulfomucins, and proteoglycans blue. Neutral mucins are stained deep red/magenta with the PAS.
PRINCIPLE AND RESULTS: This stain is intended for use by laboratory professionals to stain routinely prepared paraffin embedded tissue specimens (in vitro) to identify acid mucins, carboxylated and sulfated mucosubstances. Strongly sulfated mucosubstances are not demonstrated. Acid mucins, carboxylated and sulfated mucosubstances are stained blue.

Sample Type Required: Deparaffinized and re-hydrated tissue sections on positively charged slides. This rinse will protect the subsequent Alcian Blue Solution from pH changes due to the introduction of water. Rinse well enough to remove excess Alcian Blue.
