HIV-1 protease or PR is a retroviral aspartyl protease (retropepsin), an enzyme involved with peptide bond hydrolysis in retroviruses, that is essential for the life-cycle of HIV, the retrovirus that causes AIDS.

HIV protease, the third virally encoded enzyme, is required in this step to cleave a viral polyprotein precursor into individual mature proteins. The viral RNA and viral proteins assemble at the cell surface into new virions, which then bud from the cell and are released to infect another cell.
The gene encoding model enzyme HIV protease was randomly mutated, and the resulting library was expressed in Escherichia coli cells to eliminate cytotoxic broad-specificity variants. The surviving variants were screened for clones with activity against a reporter enzyme.
HIV protease is an aspartyl proteinase responsible for cleaving the Gag and Gag-Pol polyproteins in a late stage of the viral life cycle. Because there is no corresponding aspartyl protease that cleaves the Gag polyprotein in mammalian cells, HIV protease has been a popular target for antiretroviral drug development.
HIV-1 PR mainly targets proteins involved in three different processes: those involved in translation, those controlling cell survival, and restriction factors responsible for innate/intrinsic antiviral responses.
