It marked the beginning of the prolonged sectional conflict over the extension of slavery that led to the American Civil War.

The South, voting as a bloc on measures that challenged slaveholding interests and augmented by defections from free states with southern sympathies, was able to tally majorities. The Senate stood as the bulwark and source of the Slave Power, which required admission of slave states to the Union to preserve its national primacy. [61] [62]
Opponents claimed that Congress had no authority to prevent the introduction of a new slave state into the Union or require any other special condition for statehood that was not demanded of existing states. The House voted 82 to 78 to require the exclusion of slavery in the new state of Missouri.
Why should Missouri be any different? The vote on the amendment’s provisions brought sectional divisions into sharp relief: in the House, where northerners had the population advantage, the bill passed; in the Senate, where states had equal representation, it failed.
In the Senate the Missouri bill from the House was amended by striking out the anti-slavery proviso, 27 to 15, Otis and King voting with the minority; and with the Thomas territorial amendment inserted the bill was returned to the House for concurrence.
