16 Μαΐ 2023 · Isosorbide is used to manage anginal pain due to coronary artery disease, and it has approval for use in heart failure. The challenges of isosorbide therapy are drug interactions, adverse effects, and the development of tolerance during chronic isosorbide therapy.
Data from randomised double-blind trials show that IMD 60 mg once daily (the most widely studied dosage) has significant antianginal and anti-ischaemic effects compared with placebo after 2 weeks' treatment.
16 Μαΐ 2023 · This activity describes the indications, action, and contraindications for isosorbide as a valuable agent in treating angina pectoris due to coronary artery disease, heart failure with reduced ejection fraction, and achalasia.
10 Ιουλ 2023 · Other forms of commonly used nitrates include isosorbide dinitrate, isosorbide mononitrate, and isosorbide mononitrate sustained-release (SR). These forms are taken orally and have a longer duration of action. However, due to hepatic metabolism, the oral forms require relatively larger doses.
Thirty-seven patients with chronic, stable angina pectoris were included in a randomized, double-blind cross-over study to assess the efficacy of once- and twice-daily dosage regimens of 60 mg isosorbide-5-mononitrate, in a controlled-release formulation (5-ISMN Durules, Astra).
8 Οκτ 2016 · Plasma drug concentrations, blood pressure and heart rate were measured over 5 to 7 days following acute and repeated administrations. After the initial 30 mg dose on the first day of infarction, mean +/- standard deviation Cmax was 1237 +/- 284 nmol l-1 with a median tmax of 4 h (range 1 to 10 h).
A total of 31 patients (20 men and 11 women) with stable angina pectoris were randomized to receive either 60 mg 5-isosorbide mononitrate in a controlled release formulation once daily, or 20 mg 5-isosorbide mononitrate twice daily.
