The use of a physiological saline buffer is required for different purposes when working with NSCs, mainly for tissue or cell washing. We recommend the use of sterile commercial Dulbecco's Phosphate Buffered Saline without magnesium or calcium (DPBS).
PBS has many uses because it is isotonic and non-toxic to most cells. These uses include substance dilution and cell container rinsing. PBS with EDTA is also used to disengage attached and clumped cells. Divalent metals such as zinc, however, cannot be added as this will result in precipitation.
Phosphate Buffered Saline (PBS) is a solution prepared by diluting a stock solution of specific salts (NaCl, KCl, Na2HPO4•H2O, KH2PO4) to create a buffer with a pH of 7.3. Tween 20 is added to PBS in concentrations between 0.1% and 0.2% to optimize its performance in biological applications.
