9 Σεπ 2008 · In the first step of BER, UDG activity catalyzes uracil release and produces an abasic site in DNA. AP endonuclease produces a nick 5′ to the abasic site. The dRP lyase activity of pol β creates a 5′-phosphate and polymerase activity fills the one nucleotide gap in short-patch BER. DNA ligase seals the nick.
If dUTP:dTTP increases, DNA polymerase frequently incor-porates uracil instead of thymine during both replica-tion and repair. Uracil-DNA glycosylase removes the uracil and initiates further repair involving DNA strand breaks in an intermediate step. Repair synthesis, however, may rein-troduce uracil, leading to a futile DNA repair cycle.
9 Απρ 2021 · Here, we provide a summary of diverse uracil-DNA detection methods with their advantages and limitations, and discuss their results and conclusions. We detail the global quantitative U-detection methods as well as various emerging solutions for in situ uracil-DNA detection.

22 Σεπ 2010 · Uracil may occur in DNA due to either cytosine deamination or thymine replacing incorporation. Its quantitative characterization is important in assessing DNA damages in cells with perturbed thymidylate metabolism or within different DNA segments involved in immunoglobulin gene diversification.
16 Δεκ 2002 · Uracil in DNA results from deamination of cytosine, resulting in mutagenic U : G mispairs, and misincorporation of dUMP, which gives a less harmful U : A pair. At least four different human DNA glycosylases may remove uracil and thus generate an abasic site, which is itself cytotoxic and potentially mutagenic.
