Negative staining refers to a rapid, uncomplicated and qualitative staining technique that examines bacterial morphological characteristics. It is named negative staining because it uses negative or acidic stains that do not bind to the test specimen.

Negative staining is one of the simple stainings that can be employed for viewing bacteria by using a single stain. Negative staining employs the use of an acidic stain and due to repulsion between the negative charges of stain and bacterial surface, the stain will not penetrate in the cell.
Commonly used negative stains include uranyl acetate (UA), uranyl formate, ammonium molybdate, and phosphotungstic acid. The technique was invented by Brenner and Horne (1959) and first applied on mosaic viruses mixed with phosphotungstic acid.
